Novel ini menceritakan kisah Sitti Nurbaya dan Samsul Bahri yang sudah saling dekat sejak dari sekolah rakyat. Sitti Nurbaya diceritakan sebagai anak pedagang kaya Bagindo Sulaiman, dan Samsul Bahri adalah anak Sutan Mahmud seorang Penghulu di Padang. Keduanya harus berpisah karena Samsul Bahri harus melanjutkan sekolah dokter ke Jakarta. Novel ini juga menghadirkan sosok Datuk Maringgih yang merupakan seorang kaya yang kikir di Padang. Datuk Maringgih melakukan tipu muslihat kepada Bagindo Sulaiman (ayah Sitti Nurbaya) yang membuat ia jatuh miskin. Datuk Maringgih awalnya meminjamkan uang kepada Bagindo Sulaiman yang kemudian uang itu tidak dapat dikembalikan oleh Bagindo Sulaiman. Datuk Maringgih akhirnya mengadukan hal itu kepada Belanda agar Baginda Sulaiman dipenjarakan.
