Un elegante joven circula una tarde de otoño por Nueva York cuando una desesperada niñita reclama su atención y le suplica que entre en la lujosa mansión de su familia, pues su abuelo se ha puesto repentinamente muy enfermo y necesita ayuda. El joven, Arthur Outhwaite, se siente conmovido y acompaña a la pequeña hasta el interior de la casa, donde se convierte en la última persona que ve con vida al anciano, el gran financiero Archibald Gillespie. Antes de morir, éste le confía una carta con la petición de que la entregue a una persona concreta, y a ninguna otra. Por desgracia, la víctima fallece antes de darle más información sobre la identidad del destinatario. Cuando poco después se confirma que ha muerto envenenado, surgen infinidad de preguntas: ¿Qué contiene la carta? ¿A quién va dirigida? ¿Quién es su asesino?
